Wembley will host Super Bowl champions Philadelphia Eagles against Jacksonville Jaguars on October 28.
The fixtures, initially announced in January, will also see the Seattle Seahawks make their first trip to London when they face the Oakland Raiders at Tottenham Hotspur's new stadium on October 14.
The following week will see the Tennessee Titans take on the Los Angeles Chargers at Wembley.
The Jaguars, who will be playing in London for the sixth consecutive year, reached the AFC Championship game last season but were beaten 24-20 by the New England Patriots.
The Eagles and the Titans are also taking part in the International Series for the first time.
Confirmation of the match details comes ahead of the release of the full 2018 regular-season schedule on Thursday.
