Charlie Lee opened the scoring at London Road a minute before the interval to set Darren Ferguson's second-placed side on their way to a sixth straight league triumph.
And Chris Whelpdale was on hand to seal the success with 11 minutes remaining, although the Posh's result was partially marred by Ferguson's dismissal to the stands.
However, Peterborough, who were promoted from League Two last season, will be aware such a significant result puts them in a solid position to reach the Championship.
