Long-range strikes from Drew Talbot and Derek Niven did the damage for the Spireites despite a battling display by strugglers Town.
Grimsby fought hard all night but could do nothing about the second-half goals that condemned them to defeat.
Talbot broke the deadlock in the 55th minute when his 25-yard lob sailed over keeper Wayne Henderson and went in off the underside of the crossbar.
Henderson was beaten again when midfielder Derek Niven, who only recently recovered from testicular cancer, smashed in an unstoppable 20-yard drive in the 75th minute.
But Grimsby, who had wasted a couple of early chances, pulled one back a minute later when Ryan Bennett fired through a crowded six-yard box after Chesterfield had failed to clear a corner
