Mark Bennett will make his first Scotland appearance since winning a silver medal for Team GB at the Rio Olympics in Saturday's match against Georgia.
The Glasgow centre, who was nominated for World Rugby's Breakthrough Player of the Year award last year - returns to Vern Cotter's line-up for the clash at Kilmarnock's Rugby Park after missing the Scots' opening autumn Test clashes with Australia and Argentina.
Cotter has made five changes to the side which grabbed a last-gasp win over the Pumas at Murrayfield last week.
Bennett takes over from Huw Jones, while in the front row Ross Ford replaces Fraser Brown at hooker.
Lock Richie Gray has recovered from a head injury to take the slot filled by Grant Gilchrist, while in the back row Rob Harley and Ryan Wilson come in for Marcus Bradbury and John Barclay.
Rugby Park boasts a World Rugby approved 3G pitch and Scotland's victory over Tonga in November 2014 was the first international match involving a tier-one nation to be held on a fully-artificial surface.
Cotter said: "Our players are excited to be back in Kilmarnock and having the west country supporting us like they did two years ago will be great.
"The crowd's enthusiasm will be key to getting a good performance against Georgia. The players feed off the energy from the stands, it's one of these tangible things and really does help spur on the guys."
Meanwhile, Bennett is keen to make his mark after some impressive performances from Jones this autumn.
"I've been given an opportunity this weekend. I want to stake a claim and keep the shirt," he said.
"The competition in the squad is huge, going away to Rio when the team were in Japan meant there was the opportunity for someone else to have that chance.
"Huw was given the chance to stake a claim and that's what's happened. I need to do my best this weekend to do well and take that jersey back.
"Huw has been outstanding in the first two games, I can't say any more than that. I just need to go out and be outstanding."
